Why Melatonin Is More Than a Sleep Aid

Most people think of melatonin purely as a sleep hormone, something to take when jet lagged or restless. But melatonin is doing far more behind the scenes, and that wider role is exactly why a suppository delivery method is worth understanding.

It is made by your immune system, not just your brain. While most melatonin comes from the pineal gland, it is also synthesized directly in the bone marrow, thymus, and lymphocytes. Researcher Walter Pierpaoli, whose work on melatonin and the aging clock remains some of the most cited in the field, found that melatonin reinvigorates the thymus gland, the organ responsible for training your immune cells. A well-functioning thymus is central to fighting off chronic and stealth infections, which is part of why melatonin is so often discussed in the context of Lyme and tick-borne coinfections like Babesia and Bartonella, conditions that place a heavy, prolonged burden on immune function.

It is a potent antioxidant, not just a sedative. Independent of its sleep-regulating role, melatonin acts as a free radical scavenger throughout the body. This matters for anyone dealing with chronic illness, since oxidative stress is a major driver of the fatigue, brain fog, and inflammation that come with long-term infections.

It may help buffer the body against EMF exposure. A growing body of research has looked at melatonin’s role in reducing oxidative stress caused by electromagnetic fields, including studies on radiofrequency exposure from devices we are surrounded by daily. Melatonin appears to help neutralize the free radical activity that EMF exposure can trigger, which is one more reason many people, especially those who are highly sensitive or already immune-compromised, choose to support their melatonin levels intentionally rather than leave it to chance.

Why suppository delivery matters. Oral melatonin is broken down significantly by the liver before it ever reaches general circulation, a process called first-pass metabolism. A suppository bypasses much of that, allowing for a more direct and complete absorption, which is part of why many people find a rectal suppository more effective at the same labeled dose than a capsule or tablet.

As always, work with your practitioner to determine the dose and protocol that is right for your specific situation, particularly if you are managing Lyme, a coinfection, or another chronic condition.

Cacao butter option: Cacao butter can be substituted as the base — because it is denser than palm oil, you will need less of it. The exact amount requires some experimentation; I have not yet landed on a precise ratio. If you go this route, always verify your cacao butter is tested free of heavy metals.

Step by step instructions

  • 1Sterilize all equipment — glass or stainless steel pot, glass beaker or cup, dropper, small ramekin, and silicone molds. If your palm oil comes in plastic, transfer it to a glass jar for storage.
  • 2Gently melt palm oil in your glass pot over low heat. Do not overheat — you want it liquid but not hot.
  • 3Pour melted palm oil into your glass beaker or cup and set aside.
  • 4Place your small ramekin on the gram scale and hit tare to zero it out. Carefully weigh out 4.8 grams of melatonin powder.
  • 5Add the melatonin powder to the warm palm oil. Mix vigorously until fully and evenly incorporated — no powder clumps.
  • 6Working quickly, use the dropper from your suppository kit to fill each mold. Re-mix the liquid between fills to keep the melatonin evenly distributed.
  • 7Place filled molds on a flat plate and set on a flat surface in the freezer. Allow to solidify for a few hours.
  • 8Once solid, pop out and store in a sealed glass container in the freezer. Use one suppository in the evening before bed.
Tip: Speed matters during the filling step — palm oil begins to solidify as it cools. Have your molds ready and dropper primed before you begin filling.
